DNA脱メチル化に関わるMBD4とTDGグリコシラーゼの基質特異性

MBD4とTDGの2つのグリコシラーゼはDNAメチル化シトシンの脱メチル化に関ると考えられる。メチル化シトシンは基質にはならないが、脱アミノ化されたチミンはMBD4とTDGの基質となる。酸化されたヒドロキシメチル化シトシンは基質とならないが、さらに酸化されたホルミル化シトシン、カルボキシル化シトシンはTDGの基質となる。この基質特異性について結晶構造解析および変異体の解析から検討した。

Structural and mutation studies of two DNA demethylation related glycosylases: MBD4 and TDG

Two mammalian DNA glycosylases: the MBD4 glycosylase and the TDG glycosylase are implicated in active DNA demethylation via base excision repair pathway. 5-methylcytosine is not a substrate, but the deaminated product: thymine is a substrate of both MBD4 and TDG glycosylases. An oxidized product: 5-hydroxymethyl cytosine is not a substrate, but further oxidized products: 5-formylcytosine and 5-carboxylcytosine are substrates of TDG glycosylase. This review summarized substrate specificities of MBD4 and TDG glycosylases in view of structural studies and mutation studies.
