【Significance】
Structural flexibility provides different FTIR signals from identical CPD-photolyase repair between single-strand and double-stranded DNA

The cyclobutane pyrimidine dimer (CPD) photolyase can repair CPD type DNA damage not only in double-stranded DNA (dsDNA), but also in single-strand DNA (ssDNA). The present light-induced difference Fourier-transform infrared (FTIR) spectroscopy demonstrated largely different vibrational signals for the photorepair between dsDNA and ssDNA. However, an isotope-labeled enzyme study revealed that spectral features upon DNA repair are similar between both substrates, indicating the same mechanism. The main reason for the apparent spectral difference originates from structure flexibility of DNA after repair; ssDNA is refolded into multiple conformations, while dsDNA is refolded into a specific duplex conformation.
