【Significance】
In vitro evolution of membrane protein by liposome display

Liposome display is a newly developed technology that enables the in vitro directed evolution of membrane proteins. This technology uses cell-free translation system encapsulated inside the cell-sized liposomes, enabling the “display” of the membrane protein of interest in its functional form on the surface of the liposomes. Among the liposomes displaying different mutants, desired one could be selected by using a fluorescence-activated cell sorter. We have successfully applied this method to engineer alpha-hemolysin, a pore-forming membrane protein derived from Staphylococcus aureus. This review describes the essential characteristics of liposome display and the properties of the evolved alpha-hemolysin.
