【Significance】
Vibrational fingerprints of KcsA discriminate its interactions with alkali metal cations.

KcsA, a potassium channel exhibiting high selectivity for potassium ions over sodium ions, achieves selectivity through specific interactions between the cations and the main-chain carbonyl groups within the narrow filter region of the pore. We have applied ion-exchange-induced FTIR difference spectroscopy in attenuated total reflection mode to characterize differences among KcsA’s interactions with various alkali metal cations. Large spectral changes were caused by replacing K with Na or Li but not with Rb or Cs. The spectral similarities of the Na and Li conditions suggest that the filter forms a collapsed conformation with each of these ions.
