【Significance】
A small-angle x-ray scattering study of the disease-related mutants of human cardiac troponin

Troponin (Tn), which consists of three subunits (TnC, TnI, TnT), regulates muscle contraction via the interaction of TnT with tropomyosin (Tm). Various mutations within Tn molecules are known to cause familial cardiomyopathy. In this study, we have detected the difference in the solution structures of wild-type human cardiac Tn and cardiomyopathy-causing mutants (K247R and E244D mutations of TnT) by small-angle x-ray scattering combined with model calculations: The N- and C- terminal regions of TnT become closer to the central region of the whole molecule by the mutations, suggesting that the mutations affect the interaction between TnT and Tm.
