【Significance】
SOS/RAS positive feedback is regulated by multiple interactions between SOS and membrane components

RAS is one of the hub proteins which governs integration and divergence of various signal transduction pathways in animal cells. Activity of RAS is regulated by a positive feedback loop consisting of RAS and its activator SOS. However, the mechanism and function of the feedback loop are not fully understood especially in living cells. Here, we show that this feedback system is regulated to switch on after simultaneous interactions between SOS domains and plasma membrane components. The positive feedback is indispensable for RAS to transduce signals to the downstream RAF molecule.
